Jon Macken scored the only goal of the game as Barnsley claimed the double over their Yorkshire rivals to ease their relegation woes.
The Tykes' are fast establishing themselves as derby specialists after beating Sheffield and Doncaster twice in the last three weeks.
Macken gave Simon Davey's team a deserved lead when he slotted past Rovers 'keeper Neil Sullivan from close range after Anderson De Silva's neat pass on 42 minutes.
Jason Price looked dangerous for Rovers but wasted good chances to cap off some good approach play, while Lewis Guy's delicate lob went just wide.
